Electron Microscope Autoradiographic Study of Dopamine-synthesizing Cells in Rabbit Gastric Mucosa

Abstract
In order to study the uptake and intracellular distribution of H-dofa, a precursor of catecholamines, in the APUD cells of the rabbit gastric mucosa, 3H-dopa was injected intravenously and observed by electron microscope. One hour after injection, the pieces of rabbit gastric fundus and pylorus were removed and fixed overnight in a 2.5% glutaraldehyde solution in 0.1M cacodylate buffer (pH7.4) and post-fixed for one hour in 1.5% OSO4 in 0.3M cacodylate buffer (pH7.4). Tissues were then dehydrated in graded alcohol series and were embedded in epoxy resin.
Ultrathin sectins were affixed on a 2% celloidin coated glass slide, covered with a monolayer of ILFORD L4 emulsion by dipping method, exposed at 4¡ÆC in a light-tight box for two months, developed in microdol X, and fixed in 30% sodium hyposulfide.
Several endocrine cells of APUD series were clearly identified in the rabbitgastric fundic and pyloric mucosa by electron microscope. They were EC, ECL, X, and D cells in fundus and EC2,D, and G cells in pylorus.
One hour after injection of 3H-dopa, specific silver grains were mainly associated with the secretory granules and the number of grains in each APUD cell types were variable.
From these results, it is suggested that the endocrine cells of rabbit gastric and pyloric mucosa belong to the APUD series.
